Reconstitution Guide

A six-step laboratory procedure for reconstituting lyophilised research peptides. Adapt it to your own institutional protocols and to local safety requirements.

STEP 01

Prepare a Clean Workspace

Work on a disinfected surface in a low-traffic area. Lay everything out and within reach before you open a single stopper. Wash and dry your hands, wipe the bench down, and let the vial come up to room temperature first — usually 15 to 30 minutes out of cold storage.

  • Alcohol wipes, 70% isopropyl
  • Bacteriostatic water or your validated diluent
  • Sterile transfer syringe with a large-gauge needle
  • Marker, notebook, and a labelled sharps container
STEP 02

Calculate Your Reconstitution

Settle the working concentration before you add anything. Divide the peptide mass in the vial by the diluent volume to get mg/mL: a 10 mg vial reconstituted with 2 mL gives 5 mg/mL. Our concentration calculator will do the arithmetic for you. Write the figure down — it goes on the label and in your notes.

STEP 03

Draw the Diluent

Wipe both stoppers with a fresh alcohol wipe and let them air dry; do not fan them or blot them. Draw your calculated volume of diluent into the transfer syringe, hold the barrel up to the light, and expel any air before you go anywhere near the peptide vial.

STEP 04

Add the Diluent to the Peptide Vial

Angle the needle so the stream runs down the inner wall of the vial rather than landing on the lyophilised cake. Release the plunger slowly and let the vacuum pull the liquid in at its own pace. Directing the stream straight at the powder shears the peptide, and it is the most common avoidable loss at this stage.

STEP 05

Dissolve Without Agitation

Withdraw the empty syringe and stand the vial upright. Leave it until the cake has gone fully into solution — typically 30 to 60 seconds — and roll it slowly between your palms if it needs encouragement. Never shake it. The solution should finish clear and free of particulates; if it does not, do not use it.

STEP 06

Label and Store

Write the compound, the concentration, the reconstitution date, and the lot number on the vial before it goes anywhere. Store it upright at 2–8 °C, protected from light. Reconstituted material is typically stable for 14 to 30 days depending on the compound, so check the storage note on the relevant product page. Discard on the date you wrote, not on how the vial looks.

Good Practice

  • Aseptic technique. A fresh sterile needle and syringe for every entry into a vial, and a fresh wipe for every stopper.
  • No shaking. Vigorous agitation shears peptide chains. Slow rolling is enough, and slower is always safer.
  • Keep it cold. Vials go back to 2–8 °C straight after use. Repeated warm-up cycles cost more potency than time in storage does.
  • Document everything. Lot number, reconstitution date, concentration, and diluent — in a bound notebook or your LIMS, not on the vial alone.
  • Research use only. Nothing in this guide is intended for administration to humans or animals.
